Essay

The direct experimental evidences of the in vitro existence of non-canonical complexes EF-1alpha in GDP- and GTP-bound forms with deacylated tRNA and phenylalanyl-tRNA synthetase are presented. The Kd of [EF-1alpha*GDP*tRNA] and [EF-1alpha*GTP*tRNA] was determined. The regions of rabbit liver tRNAPhe, tRNALeu and tRNATyr involved in the interaction with EF-1alpha*GDP/GTP have been revealed. For the first time the solution conformation of rabbit liver EF-1alpha*GDP and its complex with tRNAVal has been studied by small-angle neutron scattering SANS). The model of the EF-1alpha-mediated tRNA/aa-tRNA channeling during eukaryotic protein biosynthesis was proposed.
