The dissertation is devoted to investigation of chromatin fragmentation, cytosolic and intranuclear Ca2+ concentration and nuclear Ca2+-transport in thymus and spleen lymphocytes during radiation-induced apoptosis. Using methods of DNA electrophoresis in agarose gel, investigation of Ca2+ concentration using Ca2+ fluorescent dyes fura-2 and indo-1, investigation of Ca2+-transport using 45Ca2+ and others, it has been shown that in lymphocyte nucleus exists an ATP-dependent Ca2+-transporting system. The results suggest that mechanisms of radiation-induced apoptosis in thymocytes and spleenocytes are different, besides it, the results extend our idea of nuclear Ca2+-regulation. Presented data can be used in development of methods to control lympocyte apoptotic death.
