New method of compositional fractionation of DNA has been used for study of retroviral integration in human genome in vivo. Isopycnic and compartmentalized integration of human T-cell leukemia proviruses was established for the first time. The differences in the distribution of integration sites in genomes of patients with different manifestation of disease were showed. Correlation between the quantity of integrated copies of human immunodeficiency virus in GC-poor regions of genome and the level of viral production was found. Isopycnic integration of murine and avian leukemia proviruses was demonstrated by means of compositional analysis of genes located in the neighborhood of integrated retroviral sequences. The results have provided a very consistent picture for a regional specificity of retroviral integration to be a common rule. The obtained data give a novel basic approach to the creation of a new efficient retroviral vectors for the gene therapy.
