Investigation object: isolated perfused heart and myocardium sarco- lemma preparations. Investigation purpose: finding out of a functional condition of GTP- binding proteins and phosphatidylinositol system in muscarinic acetylcholine receptors regulation of contractility of isolated perfused rabbit hearts. Investigation methods: Langendorff's method of perfusion of isolated heart, control of contractility, extraction of myocardium sarcolemma preparations, definition of GTP-binding properties of G-proteins, definition of GTPases activity, extraction of inositolphosphates, definition of protein kinases C activity, identification of substrates of myocardium sarcolemma phosphorylation. Equipment: homogeniser, electromanometer, ultracentrifuge, liquid scintilation counters. Theoretical and practical results, innovation introduced: the possibili- ty of use of myocardium sarcolemma preparations for study of mechanisms of action of physiologically active substances which have receptors on cell membrane was pro ved. The basis for development of pharmacological methods of correction of impaired myocardium contractility with failure of hormon signal transfer through muscarinic acetylcholine receptors was produced. Degree of introduction: the introduction is planned. Sphere of application: pharmacology, biochemistry, physiology.
