Molecular mechanisms of dilated cardiomyopathy (DCM) were an object of dissertation. Thesis is devoted to the investigation of functional activity and immunogenecity of cardiomyocytes' contractile apparatus regulated complex - tropomyosin and troponin at DCM in human. Methods of tropomyosin and troponin purification, protein electrophoresis, enzyme-linked immunosorbent assay (ELISA), Western-blot analysis, ion-exchange and affine chromatography, the method of reconstruction of cardiomyocites' contractile apparatus from structural (actomyosin) and regulatory (tropomyosin and troponin complex) proteins were used. The most increasing immunogenesity of troponin has been shown for DCM cardiac troponin. The changes in functional activity of troponin and the activated influence of antitroponin autoantibodies from DCM pathients' sera on actomyosin ATPase activity were revealed. The decrease in the Hsp70 level in the cytoplasm of dilated cardiomyocites was shown. Conformational changes of troponin may serve a basis of diagnostic, therapeutic and preventive measures at DCM.
