Essay

Object - the functioning of transforming growth factor ? signaling system in human lung carcinoma A549 cells treated with anticancer drugs; methods - biochemical, statistical; results - it was shown that A549 cells treated with anticancer drugs (cisplatin, doxorubicin, methotrexate) have the changes in Smad-dependent TGF?1-signaling pathway: 1) decrease in the level of mRNA expression of TGF?1 receptor type II, 2) decrease in the level of Smad2, Smad3, Smad4 proteins expression, 3) increase in the level of Smad7 protein expression which is a negative regulator in TGF?1 signaling; doxorubicin also reduced the amount of phosphorylated Smad2 and Smad3 proteins, and inhibits TGF?1-induced activation of Smad-dependent CAGA-promoter in gene reporter system; doxorubicin and cisplatin decrease the level of TGF?1-protein expression in A549 cells, but not the level of mRNA of TGF?1; introduction in Lviv national university and Lviv national medical university; branch - biology
