Landomycins are polyketide anticancer antibiotics. To study the genetic mechanisms of landomycins biosynthesis regulation. Methods of directed mutagenesis and analysis of mutants, TLC and HPLC, purification and analysis of proteins, utilization of reporter systems. Genes lndi and lanI were cloned that upregulates landomycins E and A biosynthesis in S. globisporus 1912 and S. cyanogenus S136 strains. Disruption of the genes causes a cessation of antibiotics production. Was shown that LndI bind to the structural biosynthesis genes promoter and to the promoter of its own gene. The temporal and partial pattern of lndI gene expression was studied. The role of minor bldA-tRNA in lndI gene expression regulation was investigated. The way of landomycins overproducing strains obtaining was developed. The field of application – biotechnology of antibiotics.
