Essay

The work deals with the determination of Vitamin D3 role in the mechanism of destroys of structuro-functional state of bone tissue with hypokinesia.The experiment was performed on 86 rats of Vistar line which were kept on the alimentary D-hypovitaminosis and under the condition of hypokinesia. It was showed that in the alimentary D-hypovitaminosis and the hypokinesia state, uniform metabolism changes and structuro-functional state of bone tissue took place. The metabolism and structuro-functional bone tissue changes both with D-hypovitaminosis and hypokinesia take place against a background of the content decrease of active metabolites of Vitamin D3 in the blood serum. However, the decrease of mechanisms of Vitamin D3 active metabolites in blood with the alimentary D-hypovitaminosis and hypokinesia are various. Under the conditions of alimentary D-hypovitaminosis, the cause of the content decrease in blood serum active metabolites of Vitamin is the deficit of Vitamin D3 entry into the organism that issupported by the increase of Vitamin D3-hydroxylase activity with the absence of the absorption changes of [3H]- Vitamin D3 by hepatocytes. Under the condition of hypokinesia, the cause of the content decrease of Vitamin D3 active metabolites in the blood serum is disorder of hydroxilation process of primary Vitamin D3 forms in the result of the activity inhibition of hydrolase ferments in hepatocytes that is indicated by the decrease of absorption intensity of [3H]-Vitamin D3 by hepatocytes. The data of biochemical examinations correlate with the results of histological, osteometric and histomorphometric examinations. The knowledge of the mechanism data permits to elaborate the effective pathogenetic methods of prevention and treatment of metabolism disorders and disorders of structuro-functional state of bone tissue with the alimentary D-hypovitaminosis and hypokinesia. The administration of the complex Vitamin D3 preparation (with optimal content of Vitamin D3, calcium and microelements) under the condition of experimental D-hypovitaminosis and hypokinesia favours the metabolism normalization and structuro-functional organization of bone tissue. The preparation can be recommendated for further clinical examinations in patients with disorders of structuro-functional state of bone tissue.
