Essay

The object of investigation - Ca2+-transport in mitochondria myometrium. The aim - to study of effect of spermin on Mg2+,ATP-dependent Ca2+ transport and mitochondrial permeability transition pore of smooth muscles of uterus. In the experiments, with the use of methods of a radioactive label, laser correlation spectroscopy, flow cytometry and mathematical design was gotten new information of properties of Ca2+-transporting system and mitochondrial permeability transition pore of smooth muscles of uterus, influence of spermine on Са2+ transport in these subcellular structures. It is shown, that action of spermin on Mg2+,ATP-dependent Ca2+ transport in mitochondria of myometrium is complex. Thus spermine (to 10 mM) does not influence on Mg2+,ATP-dependent Ca2+ accumulation in sarcoplasmic reticulum of myometrium. The kinetic model of the biphasic influencing of spermin is developed on initial speed of Ca2+ accumulation in mitochondria. Existence of mitochondrial permeability transition pore in myometriumis shown, which is inhibited by cyclosporine A and regulated by spermin. Results can be used in biochemical membranology. Field of application - research institution of biological and medical profile.
