Essay

The object of investigation: Wistar rats under nitric oxide hyperproduction. The aim: to investigate nitric oxide metabolism changes under it's hyper-production and possibility of their correction. Methods: electron paramagnetic resonance, spectrophotometry, statistic analysis. Formation of iron-nitrosyl complexes and EPR parameters concerning NO toxic action were investigated on following models: nitroprussid acute intoxication, subhronic nitrite natrium administration, BCG vaccination, asbestos fibers intratraheal instillation. It was experimentaly demonstrated that NO hyper-production resulted in the formation of labile ferric iron pool in blood plasma. New method to determine this kind of iron was developed. Theoretical model of NO metabolism changes correction with an exogenous NO-acceptor application was considered. The biochemical properties of the new preparation "Nitoxydel" - complex containing iron-ascorbat and unithiol were investigated. This complex can bind NO forming dinitrosyl-iron complex, carry NO and partially release NO in the organism. Methods of NO level and its toxic effect diminution and endogenous NO-synthesis intensification were developed. Field of application: research institution of biological and medical profile.
