Among the proteases involved in cell tumor transformation, the lysosomal cysteine cathepsins are in special interest. The aim of the work was the investigation of lysosomal cysteine cathepsins participation in protein degradation during carcinogenesis and the features of their endogenous inhibitors modulation in pathology. It was established that there were abnormality of lysosomal functional state that have been shown in increasing activity, changes of physical-chemical properties and intracellular compartmentalization of cysteine cathepsins B, L and H during tissue (nervous and endocrine) tumor transformation. Using author's scheme, the endogenous protein inhibitors of cysteine cathepsins from human brain without tumors and from glioblastoma were obtained and their physical-chemical properties were studied. The participation of cysteine cathepsins in degradation of fibnectin and thyroglobulin was show. Using an experimental model of postoperative somatogenic pain, the dynamics of cysteine cathepsinsin postoperative period was studied.
