Essay

The object is the chromosomal instability during the establishment of cell lines of mouse embryonic germ cells (EGCs) in vitro; the aim is to study peculiarities of chromosomal instability of mouse EGCs and the expression of Trp53 and Mgmt genes during cell adaptation to culture conditions and spontaneous neoplastic cell transformation in vitro; the methods of mammalian cells in vitro cultivation, cytology, cytogenetic, immunochemistry, biochemistry, statistical analysis were applied; the novelty lies in an obtaining and characterization of original lines from mouse EGCs. It has been shown that chromosomal instability of the G1 cell line is associated with the failure of the mitotic checkpoint, with the appearance of the mutant tumor suppressor p53 protein and with changes in expression of one of the key DNA repair enzyme - Mgmt. It was suggested a possible role of DNA repair enzyme Mgmt at the maintenance of balanced karyotype in the establishment of studied cell lines with inactivated p53; The fieldis molecular biology.
