Essay

Object - spontaneous and induced by factors of the various nature on mutational process in populations of mammalian cells; the purpose - to study features of influence various by origin and carbohydrate specificity carbohydrate binding proteins on induced mutagenesis in populations of somatic mammalian cells in vitro; the methods of mammalian cells cells in vitro-cultivation, methods of cellular biology, molecular genetics, mathematical and statistical analysis were applied; the novelty - for the first time it is shown an induction of gene mutations on a hprt locus under the influence of a number carbohydrate binding proteins of a various origin (lectins and cytokine ЕМАРП) and it is established some features of their action in population of somatic mammalian cells. Mathematical modelling of the mutational process induced by lectins, in comparison with cytokine ЕМАРП, has shown that biological action of lectins has more difficult character and it can be described the nonlinear equation of the second degree. For the first time it is revealed correlation between display of mutagenic and cytotoxic activity investigated lectins in populations of mammalian cells. For the first time it is in details studied biological activity of an animal lectin, isolated from perches caviar. It is shown that at the combined processing of cells by lectins of various origins and alkylation's agent MNNG experimentally received frequency of the induced mutations authentically more low theoretically expected size calculated under condition of their independent total action.
