Object - genetic mechanisms that control several steps of glycosylation and export of landomycins A and E in S. globisporus 1912 and S. cyanogenus S136. Goal - to determine the peculiarities of genetic control of glycosylation and export of landomycins. Methods - isolation and analysis of total and plasmid DNA, quantitative and qualitative analysis of antibiotics, purification and analysis of proteins, generation and analysis of mutations, intergeneric E. coli - Streptomyces matings, electromobility shift assays,confocal microscopy. Results and their novelty - using molecular genetic methods, the roles of genes lndW, lndJ, lanJ, lanK were determined in glycosylation and export of landomycins. Genes for late steps of L-rhodinose biosynthesis were sequenced. Proteins LndGT4 and LanZ2 were purified for the first time. Repressor protein LanK that coordinates landomycin glycosylation and efflux was extensively characterzied for the first time.
