Essay

The object - a process of development of epithelial tumours. Purpose - discovery of genetic/epigenetic changes of genes from human chromosome 3 in epithelial tumours. Methods - molecular biology methods and NotI-microarray technology. Novelty - It has been detected genes/loci of human chromosome 3 with high frequency of genetic/epigenetic changes in renal, cervical and ovary cancer in comparison with normal tissues. It have been shown for the first time that CpG-island of FHIT gene is hypermethylated in clear cell renal carcinomas and correlation exists between changes of methylation status of FHIT gene and decrease of FHIT expression. This results open possibility for using FHIT as a prognostic marker for clear cell renal carcinomas. Results - it has been found genes/loci with typical changes only for one type of tested tumours. The using of these changes can be apply for complex diagnostics of investigated tumours. It have been found seven genes/loci with high frequency of genetic/epigenetic changesin all studied types of tumours, that can reflect common mechanism of cancer development.
