Essay

The object is physicochemical properties and molecular structure of antibodies; the aim is to investigate properties and determine the primary structure of single chain antibodies (ScFv) specific to rhIFN-beta1b, which were isolated from cDNA combinatorial library of mouse variable immunoglobulin fragments; the methods are molecular-biological, microbiological, immunochemical, biochemical; the novelty lies in the fact that the panel of specific to rhIFN-beta1b ScFv's has been obtained for the first time, it has been revealed that antigen-binding center of each of the ScFv isolated has a unique structure, and the sequences themselves are new; results include obtaining of a panel of ScFv against rhIFN-beta1b, differences in the affinity constants of this antibodies as well as in their sensitivity to denaturated rhIFN-beta1b have been shown; it has been revealed that their CDRH3 regions include a conservative motif AR_P_Y with substitutions at positions 120 (F/P) and 122 (A/N), differences in the primary structure of the VL-domains have been demonstrated that are suggested to explain the discrepancy in the antigen-binding properties of the ScFv; the field is molecular-biology.
