Essay

The object - prereplicative process in the liver, induced by partial hepatectomy (PHE) and the early stage of acute phase, induced by laparatomy (LAP). The aim - studying of expression of genes Ifna1, Ifnb1 Ifnar1 and Pkr in the regenerating liver of rats. Applied methods - hepatocytes and Kupffer cells isolation; DNA and RNA isolation, RT-PCR and qRT-PCT; cloning of amplicons; Western-blot analysis and immunohistochemistry; biological test to detect fictionally active IFN alpha. Results and novelty - for the first time it has been show the up-regulation of the gene that encodes IFN alpha during first hours after PHE and down-regulation of it after unspecific extrahepatic injury (after LAP). It has been shown how Kuffer cells and hepatocytes react to up- and down-regulation of IFN alpha production after PHE and LAP, correspondingly. The expression of protein kinase R has been studied on the mRNA and protein level; changes of PKR intracellular distribution in hepatocytes were observed.
