The dissertation is dedicated to the study of functioning of the H+,K+-ATPase of rats parietal cells plasma membrane under conditions of stress- and ethanol-induced stomach ulcer. It was studies that increasing activity of H+,K+-ATPase was observed in cGMP-dependent phosphorylation at the ulcer damages, regarding to control. Analysis of kinetic parameters of purified H+,K+-ATPase had such features: decreasing of Km and increasing Vmax in ATP hydrolysis reaction, enhancing of K+-affinity and shift of enzyme pHopt to 5.0. Examination of preparation Lipin, which has an active ingredient - phosphatidylcholin it was shown normalization of H+,K+-ATPase activity at the ulcer conditions. The influence of compounds (quinoline- and pyrimidine-derivatives) on H+,K+-ATPase activity of rats parietal cells plasma membrane caused to inhibition enzyme activity. The receive results prove us that functional activity disorders of H+,K+-ATPase from stomach ulcer would be associated with significantly changes of structural organization of this enzyme, its catalytic properties and intracellular mechanism of H+,K+-ATPase activity regulation under experimental stomach ulcer development.
