Essay

The present study is dedicated to investigation of streptokinase interactions features with platelet link of the haemostatic system. Researches were conducted on the different plasminogen activation systems in vivo and in vitro. It was shown that treatment of AMI patients with streptokinase and heparin leaded to normalization of plasminogen activators inhibitor type 1 activity. On the next day after heparin therapy was canceled, the platelet aggregation degree returned to the initial high level. The activity of streptokinase was not detected in AMI patients blood flow in 1 day after thrombolytic therapy. Investigation of streptokinase influence on rabbits and porcine platelets in vivo showed that growth of PAI-1 concentration was accompanied with its activity falling. It was proved that streptokinase caused PAI-1 release from platelets in free and complex forms. PAI-1 level in cleaned platelets fraction was considerably increased in comparison with a norm in 7 days after intravenous streptokinase treatment. Investigation of immune complexes and platelets co-operation revealed that streptokinase-antistreptokinase complex caused activation of cells. It was shown that platelet aggregation degree in response to ADP was two times increased when compared to initial in 7 weeks after thrombolytic agent application. Repeated usage of streptokinase was accompanied with further growth of platelet sensitiveness to ADP.
