The present study is dedicated to the investigation of influence of proteins from Agkistrodon snake venom on the plasma and platelet links of the haemostatic system. The development of the new methogical approaches to obtaining and purification of thrombin-like enzyme, fibrinogenase, phospholipase А2, protein C activator and low molecular weight disintegrin from Agkistrodon blomhofii ussuriensis venom and plasminogen activator from Agkistrodon halys halys venom by the liquid chromatography methods were shown. The key properties of obtained proteins have been investigated. Model systems to study the influence of the proteins from Agkistrodon blomhoffii ussuriensis on platelet activation and aggregation in vitro were developed. Specificity and differential effect of the mentioned-above proteins on separate links of the haemostatic system have been established. This is the first indication that a thrombin-like enzyme purified from Agkistrodon blomhoffii ussuriensis venom has a multiple mode of action on platelets apart from its basal enzymatic activity in respect of fibrinogen. The fibrinogenase, phospholipase A2 and low-molecular weight desintegrin obtained from Agkistrodon blomhofii ussuriensis venom inhibit adrenalin- and ADP-induced platelet aggregation. For the first time it has been shown that plasminogen activator from Agkistrodon halys halys venom activates plasminogen without any additional cofactors and accelerates the proenzyme activation under the influence of streptokinase.
