Object- mechanisms of prion content regulation .Metods: biochemical, histological and immunohistochemical, cytological, bioinformatical, physicochemical and statistics. Novelty: it was estimated decrease of prion level in vitro and in vivo, in spleen and ntestine of rats by action of antisens oligodesoxyribonucleotides, complementary to the prion mRNA. For the first time the decrease of prion expression level was reached in the brain of laboratory animals via intravenous administration phosphorothioated asODNs combined with new oligoelectrolyte carriers. Oligoelectrolites could form stable nanosized supramolecullar assemblies with phosphorothioated asODNs. It is shown that the new oligoelectrolyte carriers have low cyto-and general toxicity. It was found that level of PrPC in vivo could be affected by different oligoelectrolites containing dimethylaminoethylmethacrylate.
