Tumor-associated antigens (TAA) and antibodies against them are molecular markers of cancer and can be used for diagnosis and prognosis of oncological diseases, and for studying the molecular mechanisms of carcinogenesis. Modified SEREX methodology, which consists of serological screening of cDNA expression libraries from tumors, was used to identify medullary breast carcinoma (MBC) TAA. 41 MBC autoantigens were identified 18 of which according the results of phage allogeneic screening are potential TAA of MBC. In a large-scale heterologous screening of 12 potential TAA of MBC and 4 breast cancer (BC) TAA using ELISA method 6 TAA (RAD50, PARD3, SAP30BP, SPP1, NY-CO-58, NY-BR-62) were detected, which have higher immunoreactivity in breast cancer patients. It was shown that the combination of these 6 antigens with relatively high sensitivity and specificity can differentiate healthy women and patients with breast cancer. Changes of LGALS3BP and RAD50 antigens expression were detected in breast cancer cells, and also FAM50A and PABPC4 antigens in lymphocytes infiltrating MBC that allows to consider them as potential molecular markers.
