Essay

Expression of CHI3L1 was found increased significantly in various tumors. It has been demonstrated that 293 cells stably producing CHI3L1 (293_CHI3L1) protein have the decreased doubling time and could anchorage-independent grow in soft agar. ERK1/2/MAPK pathway and AKT/PI3-kinase pathway are shown to be activated in the 293_CHI3L1 cells. 293_CHI3L1 cells stimulated the initiation of tumors after their xenograft transplantation into the adult rat brains. It was shown supression of malignant transformation in 293_CHI3L1 cells after decreasing level of CHI3L1 using specific siRNA_CHI3L1. Thus, overexpression of CHI3L1 causes malignant transformation of cells in vitro and initiates tumors in vivo suggested of CHI3L1 function in cancerogenesis.
