Essay

Object: peripheral blood erythrocytes and leukocytes of rats under normal conditions, in experimental diabetes mellitus and after treatment of agmatine. Aim: to investigate of biochemical effects of agmatine on the parameters of morpho-functional state of erythrocytes and leukocytes in experimental diabetes mellitus. Methods: flow cytofluorometry, scanning electron microscopy, immunocytochemistry, lectin-enzymatic assay, spectrophotometry, imunoblot, methods of statistical analysis.It is shown that under conditions of diabetes mellitus agmatine induses restoration of normal glucose tolerance, normalizes the intensity of erythropoiesis and restores the functional properties of red blood cells and gas transport function of blood. Agmatine demonstrates ability to reduce the intensity of peroxynitrite-mediated modification of proteins and inhibit leukocyte apoptosis. Agmatine found to normalize the sialation degree of oligosaccharide chains in glycocalyx of leukocytes and red blood cells and restore the value of the negative charge of the erythrocyte membrane that undergo changes in diabetes.
