Essay

Object: mechanisms of the growth factors biological activity. Objective: To investigate of the ability of sHB EGF to induce EGFR nuclear transportation. Methods: protein and DNA gel electrophoresis, metal affinity chromatography, protein and DNA cloning, spectrofluorometry, flow cytometry, confocal microscopy, cell culture of eukaryotes. It was shown a new mechanism of the biological activity of human sHB-EGF - its ability to stimulate the transport of EGFR into the cell nucleus. sHB EGF are found to be transported into the cell nucleus as a part of sHB EGF/EGFR ligand-receptor complex. It was shown that sHB EGF/EGFR ligand-receptor complex translocated to the nucleus via retrograde pathway. It was demonstrated that nuclear sHB EGF/EGFR ligand receptor complex had the ability to interact with the promoter region of cyclin D1 gene in vitro.
