Thesis for obtaining the scientific degree of Candidate of Sciences in chemistry in speciality 02.00.02 - Analytical chemistry. SHEE "Uzhgorod National University" МES Ukraine, Uzhgorod, 2017. Investigation of different factors such as temperature, solvent volume, and solvent flow rate on hydrophobic organic compounds recovery of from biota samples by different solvents and their mixtures during Accelerated Solvent Extraction (ASE) is described. On the base of process optimization, a rapid procedure of hydrophobic organic compounds extraction from fish muscles tissue was developed, approbated and used. The procedure provides high recovery of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) from fish muscles tissue. Extract contamination by water and water-soluble compounds is avoided. Extraction of hydrophobic organic compounds requires a small volume of organic solvents per sample. The procedure does not require expensive equipment, and can be used for OCPs and PCBs monitoring in aquatic organisms samples and for other practical tasks. A rapid micropreparative HPLC method for OCPs and PCBs isolation from aquatic organisms extracts in reverse-phase mode was developed for further determination by GC/MS. OCPs, PCBs and coplanar PCBs concentrations in fish muscle tissue samples from Dnieper river basin were determined and coplanar PCBs contribution in toxicity value was converted into 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalent. OCPs and PCBs bioavailability evaluation for aquatic organisms was done according to Сritical Tissue Level values and Bioconcentration Factor as sum of 4,4-DDTs and sum of PCBs basing on investigations results carried out in 2003 and 2011-2012.
