Essay

Object: systems of passive and active transport of Ca2+ in the human myometrium and myometrium of pig and rat. Aim: study of biochemical mechanisms of uterotonic peptide hormone oxytocin action on Ca ions homeostasis in uterus myocites. The biochemical mechanisms of the additional Ca2+ influx into myometrium cells from extracellular environment activated by oxytocin and thapsigargin were determined. It was shown that cells from nonpregnant human myometrium and PHM1-41 cells (immortalized pregnant human myometrial cells) expressed endogenous hTrpC1, 3, 4, 6 and 7 mRNA and novel splice variants for hTrpC4 and hTrpC1. The membrane-permeable derivative of diacylglycerol (OAG) stimulated an increase in oscillations of intracellular free Ca2+ concentration in PHM1-41 and myometrial cells. In the experiments which have been conducted on myometrium cell supensions of nonpregnant rats the influence of ethanol on the Ca2+ accumulation in mitochondria and sarcoplasmic reticulum have been studied.
