The principle of the immunological detection of molecular changes in tumor cells by autoantibodies and monoclonal antibody which was the main conception of this work allowed us to identify 81 autoantigens from colon, thyroid and medullary breast carcinoma tumors as well as target for monoclonal antibody MХ35 as sodium-dependent phosphate transporter NaPi2b. Characterization of immunogenicity and their expression profile allowed us to identify among them a number of tumor-associated antigens (TAAs), namely, RAD50, PARD3, SAP30BP, SPP1, NY-CO-58, NY-BR-62, LGALS3BP, BRAP, FAM50A and PABPC4. The structural and functional characteristics of antigen MХ35/NaPi2b, its gene expression profile in the tumors of patients with different cancer types and particular recognition by therapeutical antibodies MХ35 were investigated. TAAs and antigen MХ35/NaPi2b represent potential molecular markers of human malignancy, which can be used for both diagnosis and therapy of oncological diseases, and for the understanding of the molecular mechanisms of malignant transformation of cell, tumor progression and anti-tumor immune response.
