Rayevs'ka G. Obtaining biocomponents and elaboration of the high sensitive and specific ELISA test-system for the diagnostics of HIV-infection.

The work is devoted to elaborating of methodological approaches of obtaining biocomponents and developing of the high sensitive and specific ELISA test-system for the detection of HIV-specific antibodies. The HIV-specific recombinant proteins were used as antigens. The second generation ELISA test-systems were designed using anti-human immunoenzyme conjugates. The stable HRP conjugates HIV-specific recombinant proteins were obtained first and the third generation of ELISA test-system for the detection of HIV-specific antibodies was designed. The comparetive evaluation of designed second and third generation ELISA test-system has been shown the highest sensitivity and specificity of the third generation test-system. The ELISA test-kit "ІФА-ВІЛ1/2-ІІІ" was elaborated based on this third generation ELISA and the sensitivity, specificity and stability of this kit were evaluated. The ELISA test-kit "ІФА-ВІЛ1/2-ІІІ" was applied in industry at the Scientific-Productive Enterprise "Diaproph-Med" and licensedf or clinical laboratory practice in Ukraine and others countres.
