Essay

The dissertation was devoted to production and characterization of monoclonal antibodies (mAbs) against human tumor necrosis factor (TNF) with a property of mutually-enhancing or cooperative TNF binding. It has been shown that the main pair of cooperative anti-TNF mAbs consisted from competitive and allosteric neutralizing mAbs. Combination of cooperative nAbs resulted in synergistic neutralization effect and efficient TNF binding from its preformed complexes with soluble TNF receptors. Highly efficient methods for TNF detection and neutralization were developed using a pair of cooperative mAbs and their advantages in standatdization of immunoassays and neutralizing reagents were discussed.
