Cyanines with the steric hindrances, nucleic acids.Development of the approaches to obtaining of new fluorescent dyes by introduction of the substitutents in polymethine chain of trimethinecyanines and study influence of a nature of the substitutents on spectral-luminescent properties of the modified dyes at presence NA. Organic synthesis, spectroscopy, gel-electrophoresis. The C-acylation of active methylene groups of nitrogen heterocycles quaternary salts by carboxylic acids imidazolides is proposed. More than 40 dyes are synthesized and their spectral-luminescent properties in nucleic acids presence are examined. Meso-substitution of trimethinecyanine polymethine chain by primary aliphatic group decrease intrinsic dye fluorescence level, whereas meso-substituted dye fluorescence intensity in nucleic acid complex is in 2-3 times higher than intensity of unsubstituted dye complex luminescence. It was shown that the main way of trimethinecyanine-DNA interaction is intercalation. We propose highly fluorescent trimethinecyanines for use as fluorescent probes for nucleic acids detection.
