Essay

The dessertation is devoted to the use of the luminescent microscopy with acridine orange in experimental and clinical immunology. The luminescent method of determination of albumen-synthetic activity of the lymphocytes with acridine orange (AO), was adapted to the investigations of the peripheral blood, out of all indicase of the luminescence (I 530 nm; I 640 nm; ?-parameter) the most informative is I 640 nm, reflecting a level of one-chain nucleic acids (RNA) accumulated during the immunogenesis in peripheral organs of the immune system. The substitution of the autoplasma for the calf's embrio serum was proposed during preparations for the luminescent microscopy. This allows to check the luminescence of the phone. The use of the introduced indices of the luminescence (LI) allows to compare the indices of the luminescence obtained under different effort indices on photoelectronic multiplier, or on different luminescent mycroscopies. The method of smears-touches proposed, permits to estimate an albumen-synthetic activity of 20000 lymphocytes in each preparation. In an adapted state, the luminescent method with the use of AO in complex with other pathogenetic method (cytometric, ytomorphometric and avid-rosette methods), was introduced in clinical experiments. Owing to the luminescent method, leucocytal reactions on intubative and operations stress in dynamics of surgical treatment, were investigated. Migratory peculiarities of the lymphocytes of the peripheral blood in nosological groups (tumors of the central and peripheral nervous system, vertebrogenic and cranial disorders; visceropathes), were found out. The luminescent indices of the lymphocytes of the peripheral blood in oncological patients, were determined. The pathogenic significance of the luminescent method with AO was proved in comparison with lymphocytes phenotyping with the help of monoclonal antibodies (MCAB) to CD-structures (CD-3, CD-4, CD-8, CD-16, CD-25) of blood of healthy donors, and in the mitogenstimulated culture of the lymphocytes. The luminescent method with AO elaborated in a complex with other immunological methods, can be used in testing the state of the human immune system during clinical investigations.
