Essay

The dissertation is dedicated to study of "Pseudomonas batumici "- the antistaphylococcal antibiotic batumin producing strain, its taxonomic analysis, study of batumin biosynthesis, isolation, purification and characteristic of antimicrobial activity of batumin according to the international CLSI standards. The high level (99%) of genetic relatedness between "P.batumici" and P. gingeri, P. asplenii, P. fuscovaginae, P. oryzihabitans, P. putida has been shown using the partial 16S rRNA sequence analysis. At the same time the batumin-producing strain differed from these species in phenotypical characteristics which gives evidence that "P.batumici" is the new species of the genus Pseudomonas. The optimal conditions for batumin biosynthesis were: temperature 25 C; рН 7,0; the aeration coefficient 0,6 - 0,9 g O2/l h; glucose as a carbon source and urea as a nitrogen source (in the ratio C/N=10:1). The biological (diffusion in agar) and physicochemical (spectrophotometric) methods of batumin determination in the cultural liquid and in pure preparations have been developed. Two-stage column chromatography method resulted to 90% purification of batumin. Batumin antimicrobial activity against the wide spectrum of test-cultures was studied and its high and selective antistaphylococcal activity was confirmed.
