Essay

The dissertation deals with studies of condensing and water scavenging capability of chlorotrimethylsilane in reactions of carbonyl compounds. The new highly efficient methods for condensations and heterocyclizations can be applied for parallel synthesis of combinatorial libraries. The optimization studies revealed that the use of Me3SiCl in DMF insures the most efficient outcome for the condensation of carbonyl compounds leading to the formation of C-C bonds and in the heterocyclization reactions. In the case of the formation of C-N bond Me3SiCl in pyridine was proved the most efficient condensation agent. The technology was developed to carry out the condensation reactions at elevated temperatures (up to 100 C). Systematic studies revealed the scope and limitations of the combinatorial Knoevenagel procedure. Novel methodology of one-step T-effect reactions proposed. New methods for the synthesis of benzimidazoles from aldehydes and ortho-phenylenediamines was elaborated. The scope and limitations ofthis method was studied.
