Essay

The dissertation covers the studies to reversion of malignant phenotype of human lung cancer cells by long-term treatment with interferon-alpha. Objects of this study were the model of human non-small-cell lung cancer - A-549 cell line and it subline of prolonged action of IFN -alpha modification in vitro. The methods of cell culture, microscopy, immunological, molecular biology, cytogenetical and statistical are used in this study. It has been shown that long term treatment with IFN to A-549 supresses proliferative potencial of cells, the manifestation of cell's tumorigenecity in vitro, decrease expression lavel and production VEGF. It was demonstrated that prolonged action of IFN elevate sensitivity of tumor cells to cytotoxic factors chemical, physical and biological nature, and essentially alter expression level of proapoptic proteins and proteins of drug resistance. It has been shown that long-term treatment with IFN of cells changes phenotypical properties and essential changes in clonal composition of tumor cell population.
