Essay

Dissertation is devoted to the evaluation of hypoxia impact on malignant progression and assessment of clinical implication of hypoxia-associated factors for prognosis of human gastric cancer outcome. Gastric cancer tissue, surrounded "normal" mucosa, cervical cancer tissue, Lewis lung carcinoma (C57Bl/6 mice) were investigated. 31Р and 1Н NMR spectroscopy, immunohistochemical, biochemical and statistical methods were used. It was shown that 31Р NMR spectroscopy of tissue perchloric extracts is an efficient method for evaluation of hypoxia level in human tumors. It was detected that PME/Pi metabolic ratio indicates the oxygenation level of tumor, Lac/PCr ratio (1Н NMR) - glycolysis activity in neoplasia. It was shown that tumor hypoxia is a key factor of tumor progression, in particular gastric cancer. It was proposed PME/Pi and Lac/Cr ratios to use for the evaluation of oxygenation and glycolysis level in human tumors with NMR imaging (MRS).
