The dissertation was devoted to the study of streptokinase fragments manufacturing and their influence on the rabbit haemostasis system. The development of the methodological approaches to obtaining and purification of fragments by the enzyme hydrolysis and liquid chromatography methods were shown. By the methods of immune adsorption and mass-spectrometry and Edman primary structure determinations the purified fragments were identified as SK381-414 and SK1-63 and SK275-380 and SK64-380 of streptokinase Streptococcus pyogenes. In the second part of manuscript the obtained fragments influencing on the three areas of the rabbit haemostatic system was investigated. It was determined that the SK1-63 fragment, unlike the SK64-380 fragment, independently didn't influence on the fibrin-dependent or fibrin-independent plasminogen activating to plasmin. Actually by the almost unique property of SK1-63 fragment after injection, except the insignificant depressing of PAI-1 secretion, there is direct tPA releasing from endothelial cells. Important, that SK1-63 practically does not have an immunogenity. The author considers that the fragment SK1-63 can be used as a preventive at thrombotic complications.
