Essay

Investigation object - nutrient medium for cultivating the strains of Pseudomonas aeruginosa promising for biotechnology; samples of the antigens of candidates with varying degrees of acylation of protein - polysaccharides fraction eхtracted from Pseudomonas aeruginosa. Investigation purpose- the improvement of the technology of cultivating the strains of Pseudomonas aeruginosa and increase of the immunogenicity by antigens by it's succinylation. Investigation method- bacteriological, biochemical, molecular-biological, physico-chemical, mathematic-statistic. Equipment: Chromatography column for gel-chromatography with Sephadex G-75, G-200 (BioRad, Izrael); set of reagents for the determination of protein concentration, SF-56 spectrophotometer ("LOMO", Russia), Agilent 2100 bioanalyzer, Densi-La-Meter (PLIVA-Lachema as, Czech Republic). The thesis is dedicated to studying the optimal conditions of cultivating the strains of P. aeruginosa when exposed to growth-promoting factors, to investigating the immunogenicity of acylated antigen variations by the example of protein - polysaccharides fraction eхtracted from Pseudomonas aeruginosa. The possibility of substantial cheapening of the technology of obtaining antigens on account of the attrition of cultivating due to the use of low-cost raw material and enhancers was shown. The increase of immunogenicity by antigens by it's succinylation was determined. In comparison with corpuscular modified antigen, the derivative of soluble antigen acylated by 3% was twice as more efficient at immunogenicity when injection using and four times more efficient when oral using. The oral using of partialsuccinylated soluble antigen was more effective and induced antibodies level which can ensure long-term protection for an makroorganizm against P. aeruginosa. Inculcation grade: in practical bacteriological laboratories of the medical and veterinary profiles, in the science- investigation states of guard health, 1 methodical recommendation put into practice, 1 innovation.
