Spectroscopic investigations of reddened stars with observable Diffuse Interstellar Bands (DIBs) in their spectra were fulfilled in the thesis. The work is based on spectral data obtained during 2001-2009 using three spectrographs: MAESTRO, installed at the coude focus of the telescope ZEISS-2000 of the Observatory at Peak Terskol, Northern Caucasus; UVES and HARPS - spectrographs of the European Southern Observatory, Chile. Existence of 336 DIBs was confirmed and 21 new features were found. Measurements of central wavelengths, equivalent widths and full widths at the half maximum for 357 diffuse bands were fulfilled. The catalogue of the measurements and the spectral atlas of all the DIBs have been created. Profiles of relatively strong and weak DIBs were explored by high and very high resolution spectroscopy. The substructures were detected in profiles of some of them. The profiles of bands at wavelengths 585.0 and 637.9 nm at first time were explored with very high resolution R ~ 300000. The spectra were obtained with 3d camera of MAESTRO. The existence of correlation between the width of the band at wavelength 619.6 nm and excitation temperature of molecule C2 has been demonstrated. The width of this band grows and form of its profile varies in unison with the value of the excitation C2 temperature.
