Essay

The object of investigation is polyhydroxylated steroid phytohormones - brassinosteroids (BRs). Determination of BRs role in the regulation of energy homeostasis of plant cell under salinity. Methods of isotopic labeling of proteins (35S) and phospholipids (33P), methods of labeling with fluorescent phosphatidylcholine-BODIPY (phospholipid); protein extraction and electrophoresis; polarography of plant tissues, inhibitory methods for measurements of cyanide-sensitive and alternative respiratory pathways; thin-layer chromatography of plants tissue lipids; biochemical assays for enzymatic antioxidant systems activity measurements; bioinformatical methods of genes' expression analysis and proteins' identification. Using methods of radioactive labeling and methods of labeling with fluorescent phospholipid it was observed influence of BRs on early processes of formation of lipid messengers phosphatidic acid and diacylglycerol. It was found that intensity of alternative respiratory pathway increased in response to BRs action under salinity. Obtained data strongly suggests that BRs modulate synthesis of polypeptides induced by salt stress action. The application areas are bioorganic chemistry, agricultural sector.
