Essay

The work is devoted to the development of new scientifically based approaches to the selection of potentially probiotic strains of Lactobacillus and Bifidobacterium for the creation of probiotic preparations for correction of microbiota violation and indicators of immunoreactivity of the organism in cases of infectious and inflammatory diseases of the urogenital tract, which involved a comprehensive research. In the work determined the effect of Lactobacillus casei IMV B-7280, L. acidophilus IMV B-7279, L. delbrueckii subsp. bulgaricus IMV B-7281, Bifidobacterium animalis VKB and B. animalis VKL probiotic strains on the spectrum of vagina and intestine microbiota, as well as indicators of immunoreactivity of the organism in the conditions of physiological norm and staphylococcal infections of the urogenital tract in experimental animal models. For the first time made a сomparative analysis of two genetic mice lines (BALB/c and ICR) and outbred mice in indicators of qualitative and quantitative composition of the urogenital tract and intestinal contents microbiota, as well as indicators of immunity in cases of physiological norm and intravaginal infection caused by S. aureus 8325-4 strain. These data were used to create a valid model of staphylococcal infection of the urogenital tract. It is shown that all studied probiotic strains have antagonistic activity against opportunistic bacteria in vitro, and are distributed as follows according to the index of adhesiveness to epithelial cells: L. casei IMV B-7280 - B. animalis VKВ - B. animalis VKL = L. delbrueckii subsp. bulgaricus IMV B-7281 = L. acidophilus IMV B-7279, but in vivo only L. casei IMV B-7280, B. animalis VKL and B. animalis VKB strains in monoculture, as well as compositions L. casei IMV B-7280 - B. animalis VKL - B. animalis VKB and B. animalis VKL - B. animalis VKB show a pronounced antagonistic activity against coliform bacteria, microscopic fungi and S. aureus 8325-4, as well as increase the number of lactobacilli and bifidobacteria in the vagina within 30 days in cases of intravaginal or simultaneously intravaginal and oral administration. L. casei IMV B-7280, B. animalis VKL and B. animalis VKB strains (separately or in compositions) within a certain period of observation increase activity of peritoneal cavity macrophages, change phenotypic composition of spleen lymphocytes and induce the formation of IL-12 and IFN-gamma, but do not affect the content of TNF-a and IL-4, and therefore are promising for the development of probiotic preparation for the prevention of violations or correction of the vagina and intestine microbiota, as well as the immunoreactivity of the organism in cases of staphylococcal infections of the urogenital tract. It is shown that an integrated approach to the selection of potentially probiotic strains should be the basis for the creation of drugs with a broad spectrum of action, since it takes into account all mechanisms of probiotic microorganisms influence and allows to combine them correctly in the composition to enhance specific properties.
