Object - Object – biological properties of strains of M. tuberculosis isolated from patients with pulmonary tuberculosis; aim – optimization of the effectiveness of diagnosis of tuberculosis, standardizing the isolation of mycobacteria and determining their drug resistance in patients with pulmonary tuberculosis in a liquid nutrient medium in the BACTEC MGIT 960 system, with the substantiation of its combined use with new molecular genetic systems for rapid diagnosis of tuberculosis infection; methods – microbiological, immunochromatographic, molecular and genetic, statistical; results – increased the efficiency of M. tuberculosis discharge from sputum in patients with pulmonary tuberculosis in a liquid nutrient medium by using a modified dense nutrient medium to subcultivate positive samples after growing in the BACTEC MGIT 960 system; there has been proved, that the use of a liquid medium for the diagnosis of tuberculosis in children is expedient; for the first time the "critical" concentrations of second-line drugs were determined under using them for the drug sensitivity test in the Middlebrook 7H9 liquid nutrient medium using the VASTES MGIT 960 system; the standard method for the determination of MC of M. tuberculosis using a liquid medium to preparations of the second line and a reserve preparation of linezolid has been developed; there has been proved that GeneXpert molecular genetics system can be used for rapid diagnosis; the use of GenoType and GeneXpert systems for sputum examination of patients with suspected tuberculosis and patients with established clinical diagnosis depending on the clinical category of the disease has been substantiated; combined application of the GeneXpert, GenoType and VASTES MGIT 960 systems for the study of the clinical material of patients with pulmonary tuberculosis and a diagnostic algorithm for their complex use in the detection of M. tuberculosis have been substantiated; branch – medicine.
