Essay

The dissertation is devoted to the decision of the actual problem of the Ukrainian health care system - diagnosis of HIV infection in the population both in high-risk groups and in more protected social groups. It is the search for ways to develop, improve the existing domestic ELISA test systems and study the antiviral effects of potential drugs, determine their specific action is devoted to this work. A scheme for detecting HIV-1 p24 antigen in cell culture during antiviral drug research with the help of the created “DIA-HIV-Ag/Ab” test system was developed and anti-HIV activity of the biologically active substance from plant flavanoid Proteflazid was detected by the docking method the interaction of the flavonoid ring with the enzyme pocket of HIV reverse transcriptase is shown. The conditions for obtaining ICT and conjugates with biotin, stable peroxidase conjugates with original recombinant anti-HIV-1 and HIV-2 were proposed and developed. A fourth-generation immunogenic enzyme system was designed to detect antigen and HIV-specific antibodies based on these, sensitivity and specificity of this diagnostic kit were analyzed, which according to these indices is not inferior to the world analogs of fourth generation ELISA. The fourth-generation immunoassay test system for simultaneous detection of HIV1/2 antibodies and antigen p24 HIV-1 “DIA-HIV-Ag/Ab” has been introduced into production. Тhe test system was officially registered and received CE marking in accordance with the European Union standards. Testing of the qualitative characteristics of the test system was carried out at the Institute of Paul Ehrlich (Germany), at the Institute of Tropical Medicine (the Netherlands), at the Institute of Blood Transfusion Medicine and the Transfusion Medicine of the National Academy of Medical Sciences of Ukraine, AIDS Centers and blood transfusion stations.
