Object of research - influence of cryopreservation of mammalian erythrocytes using multicomponent cryopreservation media on the level of hemolysis of erythrocytes and their morphological properties. The purpose of the study is to create an effective combined cryopreservation medium for mammalian erythrocytes (bull, horse and rabbit) and to investigate the preservation of cells during the stages of cryopreservation. Research methods: differential scanning calorimetry, fluorescence microscopy, flow cytofluorometry, spectrophotometry and statistical analysis of the obtained data. For the first time, physical processes in mammalian erythrocyte suspensions with combined cryoprotective media when cooled to -196 °C were shown. It was found that the glass transition temperature of erythrocytes in combined cryoprotective media containing PEO-1500, DMSO, 1,2-PD and sucrose increases in comparison with the glass transition temperature of one-component media based on DMSO. Developed cryopreservation medium 15% PEO-1500, 10% DMSO, 5% 1.2 PD and 5% sucrose, which is highly effective in cryopreservation of erythrocytes of different mammalian species and is the most versatile of the currently studied cryoprotective solutions.
