Essay

The object of the study - the viability of isolated grape buds and in the cuttings. The purpose of the study is to create effective methods of saturation of cuttings and isolated grape buds with cryoprotective and nutrient media to increase their safety during low-temperature or hypothermic storage. Research methods: vacuum infiltration, cryopreservation of grape buds, low-temperature differential scanning calorimetry, thermal analysis, colorimetric test, cultivation of cuttings and grape buds, light microscopy, confocal microscopy, statistical analysis. New methods of saturation of cuttings and isolated buds of grapes with cryoprotective and nutrient media have been developed, their efficiency in protocols of cryopreservation and hypothermic storage of grape germplasm has been proved. For the first time, quantitative values of optimal sucrose concentrations for saturation of dormant grape buds by vacuum infiltration were determined and the viability of buds and terms of their germination depending on the degree of humidity and saturation time were studied. Low-temperature phase transitions in grape buds, which were saturated with PVS cryoprotective media by different methods, were studied.
