Essay

The object of the research – morphological and functional changes in the condition of the urinary bladder of rats with infravesical obstruction before and after administering cryoextract and conditioned media of native and cryopreserved cultures of dorsal root ganglia as well as the drug Cortexin. The goal of the research – to study the effect of biologically active compositions (cryoextract, conditioned media of native and cryopreserved cell cultures) obtained from the dorsal root ganglia as well as the drug Cortexin on the structural and functional characteristics of the urinary bladder of rats with infravesical obstruction. Research methods: modeling of infravesical obstruction in rats by applying a ligature to the urethra, obtaining a culture of satellite glial cells from the dorsal root ganglia, its cryopreservation and collection, preservation of conditioned media, obtaining and storing cryoextract of dorsal root ganglia of rats, organ bath method, histological, morphometric, immunohistochemical, biochemical analysis, methods of statistical analysis. For the first time, a positive effect of cryoextract and conditioned media of native and cryopreserved cultures of cells from dorsal root ganglia on morphological characteristics and contractile activity of the urinary bladder in rats with infravesical obstruction was established in the experimental model. For the first time the expediency of using the aforementioned biologically active compositions for the restoration of structural and functional parameters of the urinary bladder in this pathology has been experimentally proved and the intra-abdominal route of their administration has been developed. For the first time, the absence of direct organ-specific effect of the neurotrophic drug Cortexin on the structural and functional parameters of the urinary bladder in rats with infravesical obstruction was established. The effect of cryopreservation on the biological effect of dorsal root ganglia cell culture secretomes on the urinary bladder of rats with infravesical obstruction was studied for the first time. For the first time, the absence of direct nephrotoxic effects when using biologically active compositions obtained from the dorsal root ganglia in rats with infravesical obstruction was established. Field of Application: cryomedicine, medicine, biotechnology, veterinary, pharmacy.
