Essay

The dissertation is devoted to creation of a means (agents) of antimicrobial influence – pathogens of localized purulent-inflammatory processes. The practical solution of the actual problem is substantiated in the work – the technology of preparation of antimicrobial drugs on the basis of nisin is created and proved its pronounced antimicrobial activity based on the assessment of microbiological, immunological and morphological parameters. The antimicrobial action of the drugs was studied in relation to Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella and Еscherichia coli. Pure daily cultures of microorganisms were used. Colloidal silver, silver nitrate and ectericide were used as reference drugs. Analysis of the studies showed the presence of antimicrobial activity in drugs that contain nisin and silver nitrate. The remaining drugs (оther drugs) did not show bactericidal properties. On the basis of the conducted researches it was concluded that the greatest efficiency is shown by nisin, therefore, drugs based on it were selected for further research. The optimal concentration of nisin in the drug should be 0.2–0.25 g per 10 ml of mixture. Increasing this concentration is not economically feasible. The effect of modifiers on the antimicrobial action of the mixture by adding diclofenac to its composition was also studied. The studies have shown a positive effect of the combined action of nisin and diclofenac, with more inhibition observed for Pseudomonas aeruginosa, than for Staphylococcus aureus. In addition, studies have shown that the inhibition of the microorganisms grow depends more on the technology drug preparation, rather than the concentration of the active substance. In vivo studies in female VALB / cJLacSto laboratory mice were performed to determine the antimicrobial activity of the drug in living organisms. The effectiveness of the use of antimicrobial drugs based on carbopol with the active substance nisin has been experimentally proven. Laboratory mice treated with the developed drug completely recovered from infection with clinical strains within 16 days of treatment. Thus, the use of antimicrobial gel preparations based on nisin and its compositions modified with diclofenac sodium contributed to the strengthening of reparative processes, occurring in the inflammatory process of the purulent wound, as evidenced by the minimization of morphological and biochemical features of inflammatory, dyscirculatory and alternative changes.
