Essay

The thesis is devoted to: the study of structural and functional characteristics of L 929 cells during 2D and 3D cultivation; determination of optimal conditions for obtaining cell spheroids from cells of the aforementioned line, calculation of permeability coefficients and mass transfer parameters for water molecules and dimethyl sulfoxide (DMSO) cryoprotectant into spheroids of different cultivation terms (7, 14 and 21 days) at various temperatures (10, 15, 25°С), determination of optimal modes of cryopreservation for spheroids based on physical and mathematical modeling, as well as experimental testing of the determined modes according to parameters of functional activity of cells (MTT assay, ability to adhesion, proliferative and migratory activity) after spheroid thawing. The algorithm to determine the optimal cryopreservation modes proposed in our study can be used for multicellular objects of multicellular objects of spherical shape, obtained from different cell types, as well as for cryoprotective media of different composition and temperature.
