Essay

This thesis is dedicated to the development of parallel synthesis protocols leading to 3-amino-1,2,4-triazoles and 5-aminotetrazoles, with further research of biological properties of the new derivatives of these compounds. The topic of this thesis is relevant since it concerns expansion of the know n synthetically feasible chemical space of diversely substituted heterocyclic compounds, and also from the aspect of facilitation of the primary hits discovery on the early stages of the DD projects. The synthetic approaches to 3-amino-1,2,4-triazoles and 5-aminotetrazoles developed in this work are suitable for parallel chemistry – i.e, plate synthesis that does not require special conditions for the synthesis and purification for most of the combinations of starting materials. This was another requirement for the synthetic method that we have considered upon the research. Upon the validation of the proposed synthetic approaches, 176 new 3-amino-1,2,4-triazoles and 277 new 5-aminotetrazoles were synthesized. Success rate of the syntheses was 76% and 69%, correspondingly. Structure and purity of the obtained compounds was proved using NMR spectroscopy and LC/MS. For the accession of the selectivity of the binding, we have conducted analogous The practical significance of this work is 2 new synthetic approaches to 3-amino-1,2,4-triazoles and 5-aminotetrazoles that significantly expand the chemical space of the synthetically feasible molecules containing these heterocycles. In addition, the potential of the use of vast chemical spaces on the primary stages of the drug discovery projects has been shown.
