Essay

This dissertation was designed to obtain evidences for acetaminophen toxicity attenuation via modulation of its biotransformation. The ability of the "Metavit" composition being superior to methionine alone, containing methionine, some vitamins and Zn microelement, co-administered to both rats and mice intraventricularly with analgesic in toxic doses, combination of the latter with ethanol and under alcoholism to decrease acetaminophen general acute toxicity and analgesic hepatotoxicity by correcting acetaminophen metabolism in liver was shown. Analgesic toxicity was attenuated due to decrease in cytochrome P-450 2E1-dependent acetaminophen reactive species formation, increased conjugation with glutathione, and decrease in lipid peroxidation rates as well as normalization in mitochondrial respiration
